Brolin will play a man on a mission to protect his son after his wife is brutally murdered in the gritty world of Mexican drug cartels.
Penn dropped out of the role in June.
The pic will be directed by Asger Leth from Peter Craig's script with Brian Grazer producing.
Shooting kicks off in January in Mexico City.
